OPA1, a mitochondrial protein in appetite control neurons, ramps up in males on soybean oil but not in females. The same gap shows up in response to an approved obesity drug.
Male mice fed soybean oil increase production of a brain protein called OPA1 in their appetite-control neurons. Female mice do not, according to a FASEB Journal paper from researchers at Osaka Metropolitan University.
OPA1 is a mitochondrial fusion protein that helps keep a cell's energy-producing compartments intact. It sits inside a small population of neurons in the hypothalamus called MC4R neurons, the same cells setmelanotide, an approved MC4R-targeting obesity drug, is designed to activate. When researchers engineered mice to lack OPA1 in those neurons, the animals ate more soybean oil and gained more weight, with females hit harder. The drug's effect in those knockout mice also differed by sex, hinting at a molecular basis for the uneven responses patients show to MC4R-targeting therapies.
The work, led by Professor Shigenobu Matsumura, ties one protein to two reader-facing puzzles: why fatty food is hard to stop eating, and why obesity drugs do not work the same way for men and women. The caveat is the size of the case. It's one paper, in mice, on one protein, using soybean oil as the only dietary fat. Re-reports and press coverage amplify the same finding. The human translation is still a hypothesis.
